Reversal of multidrug resistance and antitumor promoting activity of 3-oxo-6?-hydroxy- ?-amyrin isolated from Pistacia integerrima

The bioactive triterpenoid 3-oxo-6-?-hydroxy-?-amyrin (1) has been isolated from multiple plant sources. In this study, chloroform fraction of Pistacia integerrima extract was processed for the isolation compound. compound identity confirmed by advanced spectroscopy technique. X-ray crystallography applied molecular structure confirmation. addition, 1 screen its activity on reversal MDR (multidrug resistance) mediated P-gp (P-glycoprotein). This accomplished using rhodamine123 exclusion multidrug-resistant human ABCB1 gene transfected mouse T-lymphoma cell line. Outcomes revealed that reversing effect comparable to verapamil as positive control in vitro. Treatment TPA-induced tumor promotion with 3-oxo-6?-hydroxy- ?-amyrin led reduction anti-tumor experiment. chemo-preventive ?- amyrin curcumin based immediate early antigen expression. Molecular docking applying Autodock Vina and i-GEMDOCK v 2.1 tools indicated gives good results, determined their fitness score specificity. Moreover, results showed precisely attached a region where co-crystallized ligand receptor previously existed. Our findings may explain use extracts an anticancer agent folk medicine.
